            <title>Pan-Amphibia distribution of the fungal parasite Batrachochytrium dendrobatidis varies with species and temperature</title>

      <abstract>This dataset provides the results of a national survey of the United States for the salamander chytrid fungus Batrachochytrium salamandrivorans (Bsal) and the amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd) conducted by the U.S. Geological Survey Amphibian Research and Monitoring Initiative from May 2014 to August 2017. Sites were sampled by capturing amphibians by hand or by traps or nets that were then swabbed individually using methods that prevent sample contamination. All swabs were then analyzed using a real-time TaqMan PCR for detection of B. salamandrivorans and Bd on the extracted DNA. The data consist of locality information and data on the individual sampled, as well as the result of the test for B. salamandrivorans and Bd. No cases of B. salamandrivorans were detected in this study.</abstract>
      <purpose>These data were collected to attempt to document the current distribution of Batrachochytrium salamandrivorans (Bsal) in the United States Locality data can be used to determine the spatial extent of this survey. No cases of B. salamandrivorans were detected in this study. The number of individuals sampled per site can be used to determine the level of sampling effort at each site. The species data provides information on the species that were sampled. We also tested the samples for Batrachochytrium dendrobatidis (Bd), another amphibian fungal pathogen.</purpose>

        <procdesc>All sampling was conducted by capturing target animals by hand, nets, or traps. After capture, we handled each animal separately using disposable powderless-vinyl gloves and new, clean plastic bags to avoid cross contamination. We rubbed rayon-tipped sterile swabs (MW-113, Medical Wire &amp; Equipment, Corsham, England) over the plantar side of one front limb, the plantar side of one hind limb, the ventral tail surface of caudates, the dorsal side of the body, and the ventral surface of the body 5 times each (Van Rooij et al 2011). We placed the swabs into sterile plastic vials with 20 μl of sterile deionized water. We recorded the snout-vent length and sex. We chilled swabs immediately after field collection and froze them at -20 C or colder. Swabs were sent frozen to the National Wildlife Health Center in Madison, Wisconsin for analysis.

We extracted DNA from swabs as described by Hyatt et al. (2007) except that 125 μl of PrepMan® Ultra Sample Preparation Reagent (Applied Biosystems, Foster City, CA) and 100 mg of zirconium/silica beads (Biospec Products, Bartlesville, OK) were used so that the entire swab was immersed during the procedure and the bead-beating steps were conducted using a FastPrep®-24 homogenizer (MP Biomedicals, Santa Ana, CA).  We used a real-time TaqMan PCR for duplex detection of B. salamandrivorans and B. dendrobatidis on the extracted DNA as described in Blooi et al. (2013, 2016). We ran reactions on the 7500 fast real-time PCR system (Applied Biosystems, Foster City, CA) using QuantiFast Probe RT-PCR mastermix kit with ROX dye (Qiagen, Valencia, CA) and BSA as per the kit instructions. We used five microliters of the PrepMan® solution containing the extracted DNA as template for the PCR. We included a negative extraction control and a standard curve run in duplicate on each PCR plate. The standard curve consisted of five different concentrations of the target sequence for B. salamandrivorans or B. dendrobatidis inserted into plasmids. The concentrations of the standards occurred at ten-fold dilutions ranging from 110-1,100,000 copies (0.5 fg – 5000 fg DNA) per reaction (on some initial runs, the standard range was 11-110,000 copies per reaction). The threshold for signal detection was set at 5% of the maximum fluorescence of the standards run for that assay. We considered a positive detection of Bsal or Bd DNA if there was a detectable signal at 40 or fewer PCR cycles and no detection in all other cases. We calculated the efficiency of each run using standard curve amplification and repeated PCR plates with an efficiency of less than 90% or greater than 110%.</procdesc>
